What is retatrutide?

Retatrutide (research code LY3437943) is a synthetic peptide developed by Eli Lilly as a next-generation incretin mimetic. Unlike semaglutide (Ozempic) or tirzepatide (Mounjaro), retatrutide acts as a triple agonist: it binds simultaneously to GLP-1, GIP and glucagon receptors.

In phase II clinical trials published in NEJM (2023), 48 weeks of retatrutide in patients with obesity led to an average weight loss of 24%. Retatrutide is currently in phase III clinical trials.
